Diesel Exhaust Is Linked To Cancer Development Via New Blood Vessel Growth

ScienceDaily (Sep. 2, 2009) — Scientists have demonstrated that the link between diesel fume exposure and cancer lies in the ability of diesel exhaust to induce the growth of new blood vessels that serve as a food supply for solid tumors.

The researchers found that in both healthy and diseased animals, more new blood vessels sprouted in mice exposed to diesel exhaust than did in mice exposed to clean, filtered air. This suggests that previous illness isn’t required to make humans susceptible to the damaging effects of the diesel exhaust.

Tumors Feel The Deadly Sting Of Nanobees

ScienceDaily (Aug. 31, 2009) — When bees sting, they pump poison into their victims. Now the toxin in bee venom has been harnessed to kill tumor cells by researchers at Washington University School of Medicine in St. Louis. The researchers attached the major component of bee venom to nano-sized spheres that they call nanobees.

In mice, nanobees delivered the bee toxin melittin to tumors while protecting other tissues from the toxin's destructive power. The mice's tumors stopped growing or shrank. The nanobees' effectiveness against cancer in the mice is reported in advance online publication Aug. 10 in the Journal of Clinical Investigation.

Cell Reproduction Research May Point To 'Off Switch' For Cancer

ScienceDaily (Aug. 24, 2009) — New insight into how human cells reproduce, published by cancer researchers at Michigan State University and the Van Andel Research Institute in Grand Rapids, could help scientists move closer to finding an “off switch” for cancer.

Cancer cells divide uncontrollably and can move from one part of the body to another. They undergo dramatic shifts in shape when they do so, said Aaron DeWard, an MSU cell and molecular biology doctoral candidate who published his research recently in the Journal of Biological Chemistry. He’s trying to figure out how certain proteins trigger cell movement and division and how cancer hijacks the system to create genomic instability.

A new approach to preventing cancer teaches the immune system to seek and destroy emerging tumors.

A cancer vaccine with a twist is making headway in clinical trials at the University of Pittsburgh School of Medicine. Rather than targeting a cancer-related virus--the way Gardasil targets human papillomavirus to prevent some cervical cancers--the new vaccine triggers the immune system to attack a faulty protein that's often abundant in colorectal cancer tissue and precancerous tissue.

Richard Nixon had every reason to be optimistic when, during his 1971 State of the Union address, he called for a concerted effort to find a cure for cancer. After all, it took only three years for the Manhattan Project to produce the world's first atomic bomb. Nixon's own presidency witnessed the 1969 moon landing, a goal set forth by John F. Kennedy in 1961.

It seemed that given enough resources there was no job that Americans couldn't tackle quickly.

But with $200 billion spent and tens of millions of cancer deaths accumulated since 1971, most would say we are losing the war on cancer. Cancer is the top killer worldwide, responsible for 7.4 million or 13 percent of all deaths annually. In America cancer will soon overtake heart disease as the top killer, claiming more than half million lives annually.

New Treatment Hope For Sufferers Of Cancer That Has Hit Patrick Swayze

A man suffering from pancreatic cancer – one of most deadly forms of the disease and which has also struck Dirty Dancing star Patrick Swayze – has been given hope of long-term survival after being treated with an advanced form of radiotherapy.

Robert Ferrant, 62, who is one of the first in Britain to undergo the procedure, was given just a few months to live after he was diagnosed with the condition.

Only 13 per cent of people who contract it are alive a year after diagnosis and only three per cent surviving for five years.

Grey Hair May Be Protecting Us From Cancer

GREY hair may be unwelcome, but the processes that produce it are now better understood and could be protecting us from cancer.

Cells called melanocytes produce the pigments that colour hair and their numbers are kept topped up by stem cells. Hair goes grey when the number of stem cells in hair follicles declines. Now Emi Nishimura of Tokyo Medical and Dental University in Japan and colleagues have found what causes this decline in mice.

When the researchers exposed mice to radiation and chemicals that harm DNA, damaged stem cells transformed permanently into melanocytes. This ultimately led to fewer melanocytes, as it meant there were fewer stem cells capable of topping up the melanocyte pool. The mice also went grey (Cell, vol 137, p 1088). Nishimura's team proposes that the same process leads to the reduction in stem cells in the follicles of older people, especially as DNA damage accumulates as we age.

Marijuana Damages DNA and May Cause Cancer

A lot of studies have shown marijuana is not good for you. It can fry the brain and contribute to psychosis. The latest one finds "convincing evidence" that marijuana smoke damages the genetic material DNA in ways that could increase the risk of cancer.

Toxic substances in tobacco smoke can damage DNA and increase the risk of lung and other cancers. However, there has been uncertainty over whether marijuana smoke has the same effect.

Scientists are especially concerned about the toxicity of acetaldehyde, present in both tobacco and marijuana. However, it has been difficult to measure DNA damage from acetaldehyde with conventional tests.

Future Of Personalized Cancer Treatment: New System Delivers RNA Into Cells

ScienceDaily (May 18, 2009) — In technology that promises to one day allow drug delivery to be tailored to an individual patient and a particular cancer tumor, researchers at the University of California, San Diego School of Medicine, have developed an efficient system for delivering siRNA into primary cells. The work is published in the May 17 in the advance online edition of Nature Biotechnology.

"RNAi has an unbelievable potential to manage cancer and treat it," said Steven Dowdy, PhD, Howard Hughes Medical Institute Investigator and professor of cellular and molecular medicine at UC San Diego School of Medicine. "While there's still a long way to go, we have successfully developed a technology that allows for siRNA drug delivery into the entire population of cells, both primary and tumor-causing, without being toxic to the cells."

One Speck Of Blood Or Tissue May Be Enough To Diagnose Cancer

A drop of blood or speck of tissue no bigger than a full stop could soon be all that is required to diagnose cancers and assess their response to treatment, research suggests.

New technology that allows cancer proteins to be analysed in tiny samples could spell the end of surgical biopsies, which involve removing lumps of tissue, often under general anaesthetic.

Researchers at Stanford University, California, have developed a machine that separates cancer-associated proteins by means of their electric charge, which varies according to modifications on the protein’s surface.

ScienceDaily (Mar. 24, 2009) — Diagnosed with an extremely aggressive form of cancer called anal sac adenocarcinoma, Oscar's future seemed bleak. Bedridden and unresponsive to chemotherapy or radiation, he would be lucky to survive three months. But thanks to an innovative new drug treatment, Oscar's cancer receded and he was walking again within two weeks.

Oscar's recovery was extraordinary enough, but his case was unusual for another reason. Oscar is a Bichon Frise, who scientists reporting in Salt Lake City, Utah at the 237th National Meeting of the American Chemical Society on March 23 call "the Miracle Dog." Joseph A. Bauer, Ph.D., and colleagues described promising results with a drug called nitrosylcobalamin (NO-Cbl) in battling cancer in Oscar and three other canines without any negative side effects. While it gives profound hope to dog owners, NO-Cbl also points to a powerful new cancer treatment for humans — one that infiltrates cancer cells like a biological Trojan horse.

Lifestyle Changes Could Prevent A Third Of Cancers: Report

An ounce of prevention is worth more than a pound of cure when it comes to cancer, according to an exhaustive international report.

The report by the American Institute for Cancer Research and the World Cancer Research Fund was released Thursday in London, England.

It calls on governments to legislate healthy living, such as:

* Mandate walking and cycling paths that encourage physical activity.
* Support policies for better-priced, healthier food choices for consumers, such as reformulating processed foods to have less sugar, salt and fat.
* Ban ads for sugary drinks and unhealthy foods aimed at children.
* Require schools to provide built-in exercise opportunities for children.

Camouflaging Of Viral DNA Could Be Crucial Step In Progression Of Cancers

ScienceDaily (Feb. 14, 2009) — An estimated 15% of cancer cases can be linked to a viral infection, however the biological changes that cause some asymptomatic carriers of a virus to develop full-blown tumors are not well understood. In a study published online in Genome Research, scientists have mapped a chemical modification of DNA in three oncogenic viruses (Epstein-Barr, human papilloma virus, and hepatitis B virus) and found that the viral genome undergoes critical changes during the progression of disease, with implications for the development of new methods of prevention, diagnosis, and treatment.

Worldwide, most people are already infected with the Epstein-Barr virus, and millions are infected with the human papilloma virus and the hepatitis B virus. Many of these individuals will develop disease, and some will eventually develop a viral-related cancer such as lymphoma, liver cancer, or cervical cancer. Understanding how infections of viruses such as these can progress to cancer in some individuals is essential to the development of new methods to attack the virus and prevent malignancies.

Scientists Find Key To Keeping Killer T Cells In Prime Shape For Fighting Infection, Cancer

Like tuning a violin to produce strong, elegant notes, researchers at The Wistar Institute have found multiple receptors on the outside of the body's killer immune system cells which they believe can be selectively targeted to keep the cells in superb infection- and disease-fighting condition.

In a study published online November 30 in Nature Immunology, the researchers describe their discovery of seven different receptors on T cells that can tamp down immune responses during a prolonged battle with an infectious pathogen or against developing cancer.

Chronic over-stimulation of the immune system can lead to poor control of infections and cancer, so the results explain why it is that these key immune cells gradually become "exhausted" and ineffective over time, says the study's lead author, E. John Wherry, Ph.D., an assistant professor in Wistar's Immunology Program.

Some Cancers Disappear Untreated, Study Finds

Cancer researchers have known for years that it was possible in rare cases for some cancers to go away on their own. There were occasional instances of melanomas and kidney cancers that just vanished. And neuroblastoma, a very rare childhood tumor, can go away without treatment.

But these were mostly seen as oddities - an unusual pediatric cancer that might not bear on common cancers of adults, a smattering of case reports of spontaneous cures. And because almost every cancer that is detected is treated, it seemed impossible even to ask what would happen if cancers were left alone.

How Eating Red Meat Can Spur Cancer Progression: New Mechanism Identified

ScienceDaily (Nov. 14, 2008) — Researchers at the University of California, San Diego School of Medicine, led by Ajit Varki, M.D., have shown a new mechanism for how human consumption of red meat and milk products could contribute to the increased risk of cancerous tumors.

Their findings, which suggest that inflammation resulting from a molecule introduced through consumption of these foods could promote tumor growth, are published online this week in advance of print publication in the Proceedings of the National Academy of Sciences (PNAS).

First Complete Cancer Genome Sequenced

Scientists decipher each of the 3 billion DNA bases from the genome of an acute myeloid leukemia tumor

For the first time, a complete cancer genome, and incidentally a complete female genome, has been decoded, scientists report online Nov. 5 in Nature. In a study made possible by faster, cheaper and more sensitive methods for sequencing DNA, the researchers pinpoint eight new genes that may cause a cell to turn cancerous.

“Since cancer is a disease of the genome, this newfound ability to determine the complete DNA sequence of a cancer cell is enormously powerful,” comments Francis Collins, a geneticist and former director of the National Human Genome Research Institute in Bethesda, Md., a group that raced to sequence the first entire human genome.

Anti-Cancer Beer Under Development

NEW YORK: American students have designed a genetically modified yeast that can ferment beer and produces the chemical resveratrol, known to offer some protection against developing cancer.

Resveratrol is a chemical found in high concentrations in grapes, berries, peanuts and pistachio nuts. It has received increasing attention since 1992, when researchers suggested that red wine containing large amounts of resveratrol might have cardiovascular health benefits.

Antioxidant effects

Mouse studies have shown that resveratrol has anti-inflammatory and antioxidant effects, and that it may stop several different stages of cancer cell development. While the benefits of resveratrol in humans remain unclear, it has become a popular health supplement.

The yeast which has been genetically modified to produce the chemical, currently contains unpalatable chemical markers, however, and is yet to be brewed into beer.

It is being developed for a student genetic engineering competition to be judged in Massachusetts, next week. Previous entries in the International Genetically Engineered Machine (iGEM) competition have included both bananas and bacterial cultures engineered to smell of mint.

The idea for the healthier beer, dubbed 'Biobeer', started out as a joke. "You could say that the inspiration for the project came from a student who really enjoys his beer," said Thomas Segall-Shapiro, a member of the team behind the project.

The team are mostly undergraduate students, based at Rice University in Houston, Texas, some of whom aren't yet old enough to legally drink alcohol in the U.S., where the limit is set at 21.

Segall-Shapiro, said that one problem with health supplements containing resveratrol is that many contain an oxidised form of the molecule, which is unlikely to be fully activated, and therefore effective, when consumed.

Cancer Stem Cells -- The Root Of All Evil?

Cancer may be caused by stem cells gone bad. If that proves to be correct, it should revolutionise treatment

MUCH of medical research is a hard slog for small reward. But, just occasionally, a finding revolutionises the field and cracks open a whole range of diseases. The discovery in the 19th century that many illnesses are caused by bacteria was one such. The unravelling of Mendelian genetics was another. It now seems likely that medical science is on the brink of a finding of equal significance. The underlying biology of that scourge of modern humanity, cancer, looks as though it is about to yield its main secret. If it does, it is possible that the headline-writer’s cliché, “a cure for cancer”, will come true over the years, just as the antibiotics that followed from the discovery of bacteria swept away previously lethal infectious diseases.

The discovery—or, rather, the hypothesis that is now being tested—is that cancers grow from stem cells in the way that healthy organs do. A stem cell is one that, when it divides, produces two unequal daughters. One remains a stem cell while the other multiplies into the sorts of cells required by its organ. This matters for cancer because, at the moment, all the cells of a tumour are seen as more or less equivalent. Therapies designed to kill them do not distinguish between them. Success is defined as eliminating as many of them as possible, so those therapies have been refined to do just that. However, if all that the therapies are doing is killing the descendants of the non-stem-cell daughters, the problem has not been eliminated. Instead of attacking the many, you have to attack the few. That means aiming at the stem cells themselves.
